The Joe Jefferson Players (JJP) opened its first production on December 17, 1947. The show was Angel Street, also famous as the film Gaslight. This first production starred Emily Bell McNally who is still active at JJP as a member of the board. JJP moved to its current location in 1949, occupying a small church building. The first show in this location was The Man Who Came to Dinner and opened November 7, 1949. The original building was eventually condemned and after operating out of the Dauphin Street Methodist Church for two years, the current building opened in 1961 with the production Look Homeward Angel. The playhouse was expanded in 1967 and has been in continuous operation ever since.

JJP was named in honor of Joe Jefferson (1829-1905), one of the premier actors in the late 1800s. Jefferson was born in Philadelphia, a fourth generation thespian. He toured with his family throughout the United States and the world. Jefferson lived in Mobile during his teenage years, and his father died here in 1842 during the yellow fever epidemic. The senior Jefferson is buried in Magnolia Cemetery on Virginia Street. Joe Jefferson was famous for his recurring role as Rip Van Winkle.
